NO there is none difference in png produce from photoshop or illustrator.
PNG is a file format, only deals for image compression, to be mainly use in network(because of it low size n good crispy output).
Now either you use windows-paint software, photoshop, illustrator, aftereffects or any of the 3d application to produce those png.
Question will have weightage if u ask, is there a differnce bw png or targa?

layering is like seggregation of an element into multi components. This is not only stick to photoshop, also illustrator, after effects, premiere n many other good softwares are having this layering feature.
