Can we do 3D Animation in flash?

If you are asking about 3D as in real mathematical 3 dimensions then its not possible. But if you want 3D as an illusion which gives a depth in your animated artworks then forget flash it is possible even in traditional animation also.

I am pretty new to zbrush and 3 d modelling. What to focus more while learning?

Try your hands in traditional sculpting with any medium available to you because on a whole its about the skill of sculpting whether you are doing it digitally or manually. Tools are nothing if you have the skills.

First of all I want to learn what do you mean by animation. If you are talking about 3D animation as a whole production process then Zbrush is really very important at the modeling, texturing stage for various technical aspects such as poly painting, color/cavity/AO/normal/ displacement map creation and many more. But if you are talking about only animation as a part of the whole production process then there is no need of zbrush.

What is the major difference between 2D and 3D Animation?

Well its a bit technical if you talk about the real difference. For 2D the animation is mostly manual like we can use a lightbox and pencil but 3D cant be done without a computer. 2D is frame by frame and 3D can be done pose to pose also. Well its a bit technical if you talk about the real difference. For 2D the animation is mostly manual like we can use a lightbox and pencil but 3D cant be done without a computer. 2D is frame by frame and 3D can be done pose to pose also. Although 3D is nothing but a developed from of 2D or lets say traditional classical animation. There are many more differences as which cant be discussed in this small box but briefly its totally on the technological side like analog vs digital kind of a thing.
